Research Area

Research Areas (Bioinorganic and Biophysical Chemistry & Health Sciences) :

  • Physical Principles of Structure, Function, and Motions of Biological Molecules: In order to confront the enigmatic issues of structure, folding, energy, motion, and function of proteins and enzymes (cytochrome c, myoglobin, lysozymes, titin I27, transferrins), biophysical chemistry research lab work interfaces physics, chemistry, and biology using advanced ultrafast kinetic (millisecond stopped flow, nanosecond laser flash photolysis), spectroscopic (NMR, UV-visible, fluorescence, Circular Dichroism (CD), double derivative FTIR etc), thermodynamic (differential scanning and isothermal titration calorimetry (DSC and ITC)), and single molecule atomic force clam spectroscopic techniques (SMFCS). Basic calculations in conjunction with kinetic, mechanical and structural modeling facilitate interpretation of the results (Journal of molecular Biology (2021), 433, 166976, Archives of Biochemistry and Biophysics (2018), 654, 146-162, Chemical Communications (2018), 54, 9635-9638, Biochemistry (ACS), 53(32) (2014) 5221-5235). 
  • Iron Metabolism and Iron Transportation: Blood plasma serum transferrin (Tf) is the principal protein involved in iron metabolism that we are studying. Tf is primarily responsible for transport and distribution of iron from the gut and liver to tissues requiring iron. For this protein, our general approach is to apply a variety of spectroscopic (UV-visible, fluorescence, Circular Dichroism (CD), FTIR), fast kinetic (millisecond stopped flow), thermodynamic (ITC and DSC), and mechanistic (single molecule atomic force clam spectroscopy) techniques to characterize the manner in which the protein environment influences the chemical and physical properties of the iron center.   •  Interplay between Metabolic disorder and Diabetes: As in subject with diabetes, the glucose concentration becomes surplus in body, which leads to glycation of several proteins in the bloodstream. Under hyperglycemia conditions, like in diabetes mellitus (DM), the glycation of blood proteins can significantly alter its ability to bind and transport metals from bloodstream to tissues.  However, the molecular mechanism by which it causes these actions is not clearly understood. The biophysical chemistry lab research work evaluates the molecular mechanism by which glycation of blood proteins modulates the structural, kinetic, thermodynamic and functional properties of blood proteins.

  • Advanced ultrafast protein folding study using laser flash photolysis and fluorescence correlation spectroscopy (FCS) in confocal microscopy.
  • Role of glycation on the mechanisms of iron and chromium binding and uptake by transferrin under hyperglycemic conditions 
  • Investigation of prevention and causes  of  protein misfolding/aggregation/amyloid fibrils formation in natively disordered proteins   
  • Structural and functional studies of metalloproteins using NMR, circular dichroism (CD), steady state and time resolved fluorescence. and millisecond stopped mixing device.   
  • Mechanical unfolding study of cardiac proteins by single-molecule atomic force spectroscopy
