Research Interests

Investigating oxysterol binding protein and lipids crosstalk in antifungal drug resistance of pathogenic fungi:

Oxysterol binding proteins are mainly involved in the transport of sterol and phosphatidylinositol 4-phosphate (PI4P) between membranes such as plasma membrane and endoplasmic reticulum, and these proteins are highly conserved from yeast to humans. Ergosterol, the main constituents of the yeast plasma membrane, influence sterol targeting drug action, such as azoles and polyenes. Amphotericin B (AmB), a polyene drug commonly used to treat fungal infection caused by pathogenic fungi Candida albicans and Cryptococcus neoformans. Recent studies have demonstrated that sphingolipid biosynthesis defective mutants also play a critical role in AmB drug resistance modulation but the exact mechanism involved is still unclear. Pathogenic fungi have been described as a global health threat due to their ability to cause invasive infections with a high mortality rate. The emergence of antifungal drug resistance in pathogenic fungi, prompt us for novel drug discoveries which are required urgently for combating pathogenicity caused by these pathogenic fungi.  We are using molecular and cell biology approaches to explore the biochemical pathways involved in lipid exchanges and drug resistance.
